Effects of Prostaglandin E1 on Cell Proliferation and Collagen Synthesis in Human Diabetic Fibroblasts: A Pilot Study

ABSTRACT
Prostaglandin E1 (PGE1) is used in diabetic foot wounds to induce healing by enhancing the blood circulation. However, the effect of PGE1 will directly have on the wound itself has not been well studied. Because whether PGE1 will have a direct positive or negative effect on wounds of diabetic patients are not known, the purpose of this pilot study is to see how, in an in vitro environment, PGE1 will affect the cell proliferation and collagen synthesis of human diabetic fibroblasts, which are major contributing factors in wound healing. In this study, diabetic fibroblasts were obtained from human diabetic patients' dermis by isolation and culture. The cells were treated with five concentrations of PGE1; 0, 3, 6, 9, and 12 ՌM respectively. After the treatment, cell proliferation was measured by Cell Counting Kit-8 assay, and collagen synthesis by Collagen type I C-terminal Propeptide method. Subsequently, the results of each group were compared. The diabetic fibroblasts treated with PGE1 showed significant increase in cell proliferation (P<0.05, Kruskal-wallis test), but showed a decrease in collagen synthesis although it may not be significant (P>0.05, Kruskal-wallis test). PGE1 stimulates cell proliferation in human diabetic fibroblasts, but on the other hand, suppresses collagen synthesis. (J Korean Wound Management Soc 2011;7:65-70)
